RhinoSeq™ DNA Library Prep builds sequencing-ready Illumina libraries in 20 to 40 minutes from as little as 10 ng of DNA, in a single tube.
Three steps in one tube
- Fragment. DNA is fragmented enzymatically, without GC bias.
- Ligate. A single master mix forms a temporary structure with sequence-specific single-stranded overhangs, so adapters ligate quickly and efficiently. The structure is degraded during the protocol and is not sequenced.
- Index and size-select. Libraries are ready to sequence, or to amplify with D5/D7 dual indexes, followed by a single size selection at >180 bp, >300 bp or dual-size.
Why it saves time
- One transfer and cleanup step, compared with three or more in traditional workflows.
- A single-tube workflow that suits automation.
- Low input, starting from 10 ng of DNA.
Choosing sizes
- Library prep kits: 8, 24 and 96 reactions.
- D5/D7 dual indexes: 8, 24 and 96 wells, sold separately.
- Applications: whole genome sequencing, with other applications in development.
